HUMINSULIN NPH is a suspension of human insulin, typically administered via subcutaneous injection (under the skin). It’s designed to provide a sustained release of insulin over an extended period, mimicking the body’s natural basal insulin production (the constant low level of insulin needed between meals and overnight).

The “NPH” in its name refers to:

  • Neutral: Indicates its neutral pH.
  • Protamine: A protein (derived from salmon sperm) that binds to insulin, forming a complex that slows its absorption.
  • Hagedorn: Refers to Hans Christian Hagedorn, the scientist who developed this formulation.

Once injected, the insulin slowly detaches from the protamine-zinc complex, allowing it to enter the bloodstream gradually. This results in an onset of action typically within 1 to 3 hours, a peak effect around 4 to 12 hours, and a total duration of action that can last up to 24 hours. Because of its extended action, it is often used once or twice daily.

It is crucial to mix HUMINSULIN NPH thoroughly before each use by gently rolling or inverting the vial/pen to ensure the insulin is uniformly cloudy or milky, as the active ingredient can settle at the bottom. It should never be injected intravenously (into a vein) or intramuscularly (into a muscle), and proper injection site rotation is essential to prevent complications like lipodystrophy (changes in fat tissue under the skin).
