Description

Deflux Tablet, containing Domperidone, is a medication used to relieve various gastrointestinal issues and related symptoms.

Uses of Deflux Tablet

Deflux Tablet is prescribed for:

• Gastritis (inflammation of the stomach lining)
• Lactation (to promote milk production)
• Migraine (for associated nausea and vomiting)
• Nausea and Vomiting
• Gastroparesis (delayed stomach emptying)
• Non-ulcer Dyspepsia (indigestion without an ulcer)

Dosage Information

Deflux Tablet is taken orally or administered rectally.

Adult Dosing

• Oral Administration:
  • Nausea and Vomiting: 10-20 mg every 4-8 hours. Maximum dose: 80 mg/day.
  • Non-ulcer Dyspepsia: 10 mg 3-4 times daily. Maximum dose: 20 mg 3-4 times daily. The last dose should be taken at bedtime.
  • Migraine (for associated symptoms): 20 mg every 4 hours. Maximum: 4 doses/24 hours.
• Rectal Administration:
  • Nausea and Vomiting: 60 mg twice daily.
• Elderly: No dosage adjustment is needed.
• Hepatic Impairment: No dosage adjustment is needed.

Child Dosing

• Oral Administration:
  • 0.2 – 0.4 mg/kg every 4 – 8 hours daily.
• Rectal Administration:
  • Children <12 years and >15 kg: 30 mg twice daily.
  • Children >12 years and >35 kg: 60 mg twice daily.

Renal Dosing

• Renal Impairment:
  • Mild to Moderate: No dosage adjustment is needed.
  • Severe: Reduce dosing to 1-2 times daily with prolonged treatment.

Administration

Deflux Tablet should be taken on an empty stomach, 15-30 minutes before meals.

Important Considerations and Warnings

Contraindications

Do not use Deflux Tablet if you have:

• Hypersensitivity (allergy) to domperidone.
• Gastrointestinal hemorrhage, obstruction, or perforation.
• A prolactin-releasing pituitary hormone (prolactinoma).
• A need for chronic administration or routine prophylaxis of postoperative nausea and vomiting.

Precautions

Use with caution in patients with:

• Phaeochromocytoma (a tumor of the adrenal gland).
• Children under 2 years old.
• Elderly patients.
• Renal or hepatic impairment.
• Risk of cardiac arrhythmias and hypokalemia (low potassium) if administered intravenously (IV).

• Dysrhythmias in patients with cardiovascular disease or hypokalemia
• Seizures
• Hypertensive crisis in patients with phaeochromocytoma
• Cardiac arrest (in cancer chemotherapy patients)

Mechanism of Action

Domperidone acts as a peripheral dopamine-receptor blocker. It works by:

• Increasing esophageal peristalsis (wave-like contractions).
• Enhancing gastroduodenal coordination.
• Lowering esophageal sphincter pressure.
• Increasing gastric motility and peristalsis.

These actions facilitate gastric emptying and decrease small bowel transit time, helping to move food through the digestive system more effectively.
